The diarrhea observed in patients which cholera is known to be related to secretion of water and electrolytes into the intestinal lumen. However, the exact mechanisms involved in these secretory processes have remained unclear. Although it is clear that purified toxin acts on epithelial cell metabolism, its activity on Na+ transport across intestinal mucosa is equivocal: reported either to prevent net Na+ absorption or to cause net secretion of Na+ from serosa to mucosa. Since total transmural Na+ fluxes across “leaky” epithelia involve very significant movement via a paracellular shunt pathway, we studied the effects of cholera toxin on the cellular and paracellular pathways of Na+ movement. Unidirectional Na+ fluxes were examined as functions of applied potential in control tissues and in tissues from the same animal treated with purified cholera toxin. Treatment of rabbit ileum in vitro with toxin stimulated the cellular component of serosa-to-mucosa Na+ flux (from per cm2 under control conditions to per cm2 after treatment with toxin, ). The effect of cholera toxin on Na+ movement through the cells from mucosa to serosa appeared to be insignificant. Finally, a marked decrease in the Na+ permeability () and no detectable significant changes in transference number for Na+ of the paracellular shunt pathway were observed following treatment with cholera toxin. These results provide direct evidence for the hypothesis that purified cholera toxin stimulates active sodium secretion but has minimal effect on sodium absorption. 相似文献

Intraflagellar transport (IFT) machinery mediates the bidirectional movement of cargos that are required for the assembly and maintenance of cilia. However, little is known about how IFT is regulated in vivo. In this study, we show that the small guanosine triphosphatase (GTPase) adenosine diphosphate ribosylation factor–like protein 13 (ARL-13) encoded by the Caenorhabditis elegans homologue of the human Joubert syndrome causal gene ARL13B, localizes exclusively to the doublet segment of the cilium. arl-13 mutants have shortened cilia with various ultrastructural deformities and a disrupted association between IFT subcomplexes A and B. Intriguingly, depletion of ARL-3, another ciliary small GTPase, partially suppresses ciliogenesis defects in arl-13 mutants by indirectly restoring binding between IFT subcomplexes A and B. Rescue of arl-13 mutants by ARL-3 depletion is mediated by an HDAC6 deacetylase-dependent pathway. Thus, we propose that two conserved small GTPases, ARL-13 and ARL-3, coordinate to regulate IFT and that perturbing this balance results in cilia deformation. 相似文献

Selection and isolation of bacteria capable of degrading dinoseb (2-sec-buty-4,6-dinitrophenol) 总被引:3,自引:0,他引:3
Dinoseb (2-sec-butyl-4,6-dinitrophenol) has been a widely used herbicide that persists in some contaminated soils, and has been found in groundwaters, causing health and environmental hazards. Persistence in some soils may stem from a lack of dinoseb-degrading organisms. We established a chemostat environment that was strongly selective for aerobic (liquid phase) and anaerobic (sediment phase) bacteria able to degrade dinoseb. The chemostat yielded five taxonomically diverse aerobic isolates that could transform dinoseb to reduced products under microaerophilic or denitrifying conditions, but these organisms were unable to degrade the entire dinoseb molecule, and the transformed products formed multimeric material. The chemostat also yielded an anaerobic consortium of bacteria that could completely degrade dinoseb to acetate and CO2 when the Eh of the medium was less than-200 mV. The consortium contained at least three morphologically different bacterial species. HPLC analysis indicated that dinoseb was degraded sequentially via several as yet unidentified products. Degradation of these intermediates was inhibited by addition of bromoethane sulfonic acid. GC-MS analysis of metabolites in culture medium suggested that regiospecific attacks occurred non-sequentially on both the nitro groups and the side-chain of dinoseb. The consortium was also able to degrade 4,6-dinitro-o-cresol, 3,5-dinitrobenzoic acid, 2,4-dinitrotoluene, and 2,6-dinitrotoluene via a similar series of intermediate products. The consortium was not able to degrade 2,4-dinitrophenol. To our knowledge, this is the first report of strictly anaerobic biodegradation of an aromatic compound containing a multicarbon, saturated hydrocarbon side chain.Abbreviations BESA
